A party of 6 or so should be able to clear this, barring horrible luck. Typical setup would be NIN, 2 other blinked melees, RDM BRD BLM. WHM might be nice as well because of the lizard floor.
The chests drop randomly from mobs, a single floor is not necessarily going to contain a time, restore, and treasure chest. You can open as many as you find, you'll likely get a few restore chests, so use your two hours often (provided they're helpful...don't Mijin Gakure repeatedly...). Open them as you find them, the restore chest being the possible exception if you don't need it at the moment and there are still mobs left on the floor.
Killing a single mob on the floor will open the vortex to the next floor, so keep this in mind along with what floor you are on and how much time you have left.
Probably best to kill one slime and then move on unless there are 2 or more BLMs.
Mages be careful with the Mandy floor, time is crucial when going in small groups and getting slept wastes a significant amount of time.
